Central-acting melanocortin peptide studied for desire and arousal.
PT-141 — bremelanotide, a melanocortin peptide studied in sexual desire and arousal research.
PT-141, also known as bremelanotide, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone (α-MSH) that acts as an agonist at melanocortin receptors MC3R and MC4R in the central nervous system — specifically the hypothalamus and limbic system. Bremelanotide is approved in the United States under the brand name Vyleesi for a specific type of acquired, generalised hypoactive sexual desire disorder (HSDD) in certain premenopausal women.

PT-141 activates melanocortin receptors, including MC4R, in the central nervous system, influencing brain pathways associated with sexual desire and modulating signalling involved in sexual motivation and arousal. Unlike PDE5 inhibitors such as sildenafil and tadalafil, which act peripherally on vascular smooth muscle, it operates centrally on the neural circuits governing sexual function.
FDA approval followed in 2019, when Vyleesi was licensed for hypoactive sexual desire disorder in premenopausal women, making bremelanotide one of the few peptides with full FDA approval. Research has demonstrated efficacy in both male and female sexual dysfunction, with effects mediated through dopaminergic and oxytocinergic pathways downstream of MC4R activation.
Bremelanotide is a prescription medicine in the United States and is approved only for a specific diagnosis in certain premenopausal women. A medical assessment is advised before considering any compound that acts on the central nervous system; ask a qualified professional to review your medicines and medical history first.
Its effectiveness is not established for every type of sexual dysfunction, for postmenopausal women, or for improving sexual performance in people without a diagnosed condition.
No self-directed cycle is recommended. Bremelanotide is approved only for a specific diagnosis, so a qualified professional should confirm suitability, dose and interaction checks before any use.
Reconstitution: add 3 ml of bacteriostatic or sterile water. Use sterile single-use equipment and needles, do not share a cartridge, and ask a qualified professional to review the intended route and handling before use.
Follow the manufacturer’s label for the exact formulation, temperature range and expiry before and after opening or mixing. Request these instructions before ordering if missing. Do not assume a universal 30-day shelf life; keep away from heat and light and out of children’s reach. Ask for advice after any temperature excursion.
Bremelanotide is approved for one specific diagnosis; every other use remains under study.
Do not self-stack research compounds or hormone-active products. Ask a qualified clinician to review ingredients, medicines, medical history and monitoring requirements.
